In cultured chick retinal pigment epithelium, vinculin (a 130,00 dalton actin binding protein) was demonstrated using biochemical and immunohistochemical methods. Vinculin was shown to be localized at the adherent plaques of the retinal pigment epithelial cells at the early phase of cell culture. As the stage of the culture progressed, the intracellular distribution of vinculin was changed drastically to the lateral cell membrane. Actin was also demonstrated to change its distribution in accordance with the distribution change of vinculin. The possible role of vinculin and actin in the process of migration of retinal pigment epithelial cells and in the development of cellular polarity was discussed.
