Changes in body and adrenal weights, adrenal cholesterol content, plasma corticosterone concentration as well as in nuclear volumes and mitotic index in zona fasciculata of the adrenal cortex were studied in rats during 12 hours and 1, 3, 5, 7, 10, 20, 30, 45 and 65 days of the readaptation period after 20 days of movement restrain in pencil-cases. The removal of rats from pencil-cases and return to usual conditions caused the development of readaptation stress lasting 7 days. When the signs of the stress in posthypokinetic rats disappeared, some damages concealed before were found: retardation in age growth of adrenals, retardation in age growth of cell nuclei in zona fasciculata, a decrease in the basal level of cholesterol in adrenals, which could result from total catabolic changes induced by hypokinesia. In cells of adrenal zone fasciculata an increase of mitotic division and then hypertrophy of nuclei were observed; these changes could be attributed to the development of recovery processes replenishing the tissue deficit resulted from retardation in the age growth of adrenals.
