Role of prostaglandins in pentylene tetrazole induced convulsions in mice

Slow i.v. infusion of penthylenetetrazole (PTZ) resulted in clonic convulsions followed by tonic extension of hind limbs and death. Strong cyclooxygenase inhibitors, namely indomethacin, ibuprofen and sodium meclofenamate, increased the convulsive threshold, but weak cyclooxygenase inhibitors like paracetamol and eicosatetraenoic acid (ETYA) were without significant effect. Piroxicam, an inhibitor of endoperoxide isomerase and synthetase, failed to affect PTZ convulsive threshold dazoxiben, a thromboxane synthetase inhibitor, inhibitor, increased it. In addition, leukotriene receptor antagonist, FPL55712 [7,[3-(4-acetyl-3-hydroxy-2-propylphenoxy)-2-hydroxypropoxy]-4-oxo-8-propyl-4H-1-benzopyran-2-carboxylic acid] reduced the convulsive threshold. Increase in thromboxane A2 (TxA2) in CNS following slow i.v. infusion of PTZ promotes convulsions.
