Immunohistochemical demonstration of ceruloplasmin in the livers of Indian childhood cirrhosis

An immunoperoxidase procedure was used to identify ceruloplasmin in 15 liver biopsy specimens from patients with Indian childhood cirrhosis (ICC) to find its staining characters and to investigate if it differed from normal age matched controls, neonatal liver or other chronic childhood liver disorders. A relationship between immunohistochemical staining and histochemical copper as demonstrated by Rhodanine was determined. Diffuse intense brown cytoplasmic staining for ceruloplasmin present in all the ICC patients did not differ from that in the liver biopsies of the neonates or from other childhood liver disorders. Both Cu and Cu associated protein (CAP) were present in all the ICC patients. In 2 patients of ICC, rhodanine staining revealed a diffuse red cytoplasmic staining of Cu besides the typical red granules in the hepatocytes. The hepatocytes were darkly stained for ceruloplasmin in these patients.
