Participation of cell surface anionic sites in the interaction between Trypanosoma cruzi and macrophages

Incubation of bloodstream trypomastigotes of T. cruzi with cationized ferritin before interaction with macrophages significantly reduced their surface charge and markedly increased their ingestion by macrophages. Similar treatment of epimastigotes had no effect on surface charge, nor on ingestion by macrophages. Incubation of macrophages with cationized ferritin before interaction with T. cruzi markedly reduced the ingestion of epimastigotes and increased the ingestion of trypomastigotes. Macrophages labelled at 4 degrees C with cationized ferritin and then incubated with T. cruzi at 37 degrees C interiorized the anionic sites of the cell surface into endocytic vacuoles. However, no cationized ferritin particles were seen in association with the membrane of endocytic vacuoles containing parasites. Macrophages were also incubated at 37 degrees C in the presence of cationized ferritin or horseradish peroxidase. It is considered that during interaction, cell surface anionic sites of both cells and parasites are involved.
