Effect of histamine H-2 blocker - metiamide - on the isolated atrial preparations of rabbits & guinea pigs

Cardiac effects of metiamide - a histamine 2 blocker - was studied in isolated cardiac tissue preparations of rabbits and guinea pigs. The force of contraction and atrial rate were significantly increased in normal atria with increasing concentrations of metiamide; this effect was significantly decreased in reserpinized and propranolol treated tissues. A greater sensitivity to cardiac effects of metiamide was seen in guinea pigs. The dose dependent cardiostimulant effect of metiamide may be an indirect effect probably mediated through release of catecholamines from adrenergic neurons in the atria.
