Determination of desipramine by high-performance liquid chromatography in the blood and brain of mice

A new high performance liquid chromatography method for determination of desipramine blood and brain concentrations in mice is presented. The technique was developed on a normal phase column, .mu. porasil Waters using isocratic elution and UV detection at 222 nm. The solvent mixture is diehtylamine-water-acetonitrile-methanol. The mouse blood sample is extracted with hexan at a pH 11. The brain sample is first homogenated in acetic acid (0.1 N) and then extracted in hexan. The use of an internal standard allows a better reproductibility. The samll volume of blood required and the high sensitivity of this method gives a tool to study correlation between blood and brain concentrations in the same mice.
