The ultrastructural appearance of tissues and organisms varies according to the fixation used; this is demonstrated using C. albicans. Aldehyde alone as a fixative preserves the general structure of the cell for subsequent cytochemical procedures, etc., but specimen contrast is low. Potassium permanganate demonstrates membrane systems well, but not ribosomes or lipid; osmium tetroxide demonstrates lipid and ribosomes. These factors should be considered when setting up a routine processing schedule.
