The effect of heparin pretreatment on the liver before endotoxin administration in vivo and the effect of endotoxin on liver parenchymal cells in an organ (liver slices) culture were studied. Heparin pretreatment before endotoxin injection prevented liver parenchymal cells from degenerative and the necrotic changes. Heparin pretreatment was not effective on the initial (30 min to 2 h) aggregations of platelets, neutrophils, Kupffer cells, etc. in the sinusoids. A direct effect of endotoxin on liver parenchymal cells was not observed in the organ culture. The change of liver parenchymal cells after endotoxin injection apparently was not induced by the direct action of endotoxin, but was probably due to the formations of cell-aggregates, especially thrombi. Heparin pretreatment induced a slight thrombi-formation, but could not inhibit the aggregation of platelets around Kupffer cell.
