A novel growth factor for mouse myeloid leukemic cells: partial purification and characterization

A growth factor which promotes the multiplication of mouse myeloid leukemic cells (MI) was partially purified from mouse serum. The purification procedure included affinity chromatography using Blue Sepharose CL-6B and gel filtration on Bio-Gel P-150. Although the factor was co-fractionated with albumin upon the gel filtration, 71% of its activity was separated from albumin by the affinity column. The purified factor had a MW of about 65,000 daltons as determined by gel filtration at pH 7.4. Its isoelectric point was .apprx. 5.5. It was stable to heat (70.degree. C for 30 min) and acid (pH 2.3) treatment, but was degraded by pronase. The dissociation at an acidic pH of the factor into a low MW form was not observed. Comparison of several properties of the factor with other established growth promoting serum factors suggests that it is a novel growth factor.
