ECA (Enterobacteriaceae common antigen: Kunin's antigen) preparation and Salmonella typhi O antigen were tested for serological activity by passive hemagglutination, hemagglutination inhibition, precipitation and immunoelectroprecipitation. The effect of phospholipase A and NaOH on serological activity of these preparations was tested. The lowest amount of ECA needed for coating 1 ml of a 10% erythrocyte suspension was 50 .mu.g/ml. Hemagglutination inhibition with Escherichia coli O14 antiserum was obtained with 0.78 .mu.g/ml of the preparation. Agar gel precipitation with E. coli antiserum was obtained with ECA concentrations up to 62 .mu.g/ml. Phospholipase A and NaOH at 100.degree. C induced a loss of the Kunin's antigen capability of coating erythrocytes and inhibited passive hemagglutination.
