Use of leukocyte adherence inhibition test to study the cell mediated antitumour immunity in cancer patients

Nineteen patients of oropharyngeal cancer, 8 patients of breast cancer and 7 patients of carcinoma of cervix were investigated for cellular sensitization to specific tumor-associated antigens by modified leukocyte adherence inhibition (LAI) assay. As controls, leukocytes from normal individuals were tested for LAI with tumor extracts under investigation and normal tissue or leukocyte extracts. A baseline for the per cent inhibition of adherence of normal leukocytes, derived from these experiments, was approximately 20%. Leukocytes obtained from patients, mostly in pre-operative stages, were tested with autochthonous tumor extracts and allogeneic extracts from tumors of same histologic type as specific antigens. As non-specific antigens, extracts from unrelated tumors or normal tissues were tested. All 19 patients with oral cancer, 6 of the 8 patients with breast cancer and 6 of the 7 patients with cervical cancer showed increased LAI in the presence of specific antigens. Two patients of oral cancer, 1 each of breast cancer and cervical carcinoma, reacted to non-specific antigens. LAI was a reliable and reproducible assay for CMI [cell-mediated immunity], which could be undertaken under modest laboratory conditions.
