In the work changes are described in a number of adherent cells depending on several basic methodic factors, which may be important during the performance of the leukocyte adherence inhibition test, as follows: heparin concentration in the blood sampled, time between sampling the blood and leukocyte separation, laboratory glass material, sedimentation medium, method of performing the hemolysis during the leukoctye separation and composition of the medium used. The most important factor affecting the number of adherent cells was the composition of medium used for the separation and incubation of the cellular suspension and relative representation of particular species of leukocytes in the donor's blood. Changes of the concentration of Ca2+ and SO2- ions in medium and changes of the relative number of lymphocytes in the donor's blood resulted in 70% of the spread in the adherent cells portion.
