The effect of pentagastrin and secretin on the cell renewal and structure of the jejunal mucosa was studied in rats after 2 days of fasting. Pentagastrin (175 .mu.g/100 g) or secretin (24.5 CU/100 g) were injected during the 2nd day of fasting, the total dose being divided in repeated i.p. injections. Control rats received the same volume of saline solution. The animals were sacrificed during the following 24 h. The fasting lowered significantly the labeling index, wet weight and thickness of the intestine. After pentagastrin injections (24 h), the labeling and mitotic indices were significantly higher than in controls, the villus serosal area and the thickness of the muscular layer were increased. Secretin injections induced an early decrease of the labeling index followed by a decrease of the mitotic index; the number of short villi increased. These results confirm the trophic effect of gastrin on cell renewal and the 3-dimensional structure of the jejunum and indicate that secretin is an antitrophic factor.
