Influence of cyclophosphamide on the antibody response evoked by an experimental sarcoma in rats

Cyclophosphamide has been used in tumor treatment and in post-transplantation immunosuppression. An in vitro 51Cr test system, which allows studies of serum-mediated immune responses, was used to evaluate the effect of cyclophosphamide in tumor-bearing rats. A test system to evaluate the influence of antineoplastic drugs on the antibody-mediated immune response evoked by an experimental rat sarcoma is presented. Serum samples from PW-13 tumor-bearing Wistar-Furth rats were added to monolayers of 51Cr-labeled tumor cells. A clear cytotoxic effect was noted. In vivo cyclophosphamide treatment of rats after tumor inoculation virtually prevented the appearance of such antibodies, although at the same time a decreased in vivo tumor growth rate was recorded.
