Currently employed methods of differential assay of hormones and autacoids in mixtures are inaccurate, e.g., the method of assaying prostaglandins . A new principle of differential assay of autacoids is proposed based on the introduction into Vane's cascade of absorption columns which selectively remove biologically active substances interfering in the assay of selected autacoids. The principle of differential assay of autacoids was applied in the biological assay of PG produced by blood vessels under the influence of high concentrations of catecholamines, and for assay of histamine in the presence of PGE2, RCS and SRS-A released from isolated lungs of sensitized guinea pigs during hypersensitivity shock. Use of the column-cascade method permitted basic studies on the role of PG produced in the respiratory system, differentiation of the influence of steroid and nonsteroid anti-inflammatory drugs on release of mediators of anaphylactic shock and an elucidation of the antiregulative role of PG in vasoconstriction.
