Neuromuscular blocking activity of a new series of semisynthetic quaternary pyrrolizidines

A new series of semisynthetic quaternary pyrrolizidines based on pyrrolizidine alkaloids obtained from plant source . These compounds showed varying degrees of neuromuscular blocking activity resembling that of curare. The ED50 values of these compounds were determined and structure activity relationships elucidated. The derivatives of monocrotaline and retronecine having unsaturated necine base were hepatotoxic and were also far less effective as compared with those of 1-methylene pyrrolizidine and heliotridane, which have a saturated necine base and were non-hepatotoxic.
