Universal medium for extending the useful life of spermatozoa in vitro

The suitability of a universal medium (see US Patent 3 816 249 (1974)) for diluting semen was evaluated in relation to sperm characters in semen diluted with physiological saline, Lake's diluent or Minnesota buffer and in whole semen. Trials with bull and turkey spermatozoa are reported. Data are tabulated for the motility of turkey semen stored at 5 deg -6 deg C for up to 24 h. Bull semen which had been diluted with the universal medium before freezing yielded 25-55% motile spermatozoa after thawing, and use of this semen resulted in CRs of 75-80%. The fertility of turkey hens inseminated with tom semen which had been diluted with the universal medium and stored at 5 deg averaged 88.4%.
