Pharmacological tests in cancer chemotherapy

1) From the example of 7 different alkylating substances it is tried to find quantitative parameters for the characterization of their chemotherapeutic value in rat tumors. 2) For the comparison of the effectiveness of different cancerotoxic substances it is recommended not to state the curative doses only absolutely in mg/kg but also relatively in per cent for instance of the medium lethal dose. 3) The quantitative determination of the dose-response-relationships yielded in the probit-net linear regressions. 4) As the determinative parameter for pharmacological actions owns the dimension of a (reciprocal) time, the temporal course for the curative as well as for the toxic action is determined. The optimum is reached, when the toxic action is quickly reversible and when the curative action cumulates. 5) For the increase of the therapeutic range the principle is recommended to bring the substance, by a chemical linkage to a carder molecule, into an inactive « transport form out of which the active « action form » is liberated only in the organism. 6) From the example of a cyclic Alcanolamide-(bisi3chloroethyl)-phosphamide-compound it is showed that on this way it is possible to receive well tolerated and highly active cancerotoxic substances. 7) The best results promises the combination of surgery with chemotherapy. After experimental experiences it is recommended to carryout operations on cancer under « chemotherapeutic protection ».
