Although throughout the years emetine has been used in a variety of parasitic diseases other than amebiasis, including Chagas' disease, no laboratory testing against Trypanosoma cruzi has previously been reported. Reported here are the results of studies on emetine, and several related compounds and moieties thereof, in experimental Chagas' disease in mice. Cephaeline, dehydroemetine, and six dimethoxy isoquinolines were without action in this infection. The activity of emetine against T. cruzi could usually be demonstrated when parenteral doses bordering on the toxic were given. The degree of activity obtained did not suggest that it would be a useful agent for clinical treatment of Chagas' disease. Substitutions on the nitrogen in the isoquinoline moiety of emetine reduced activity as well as toxicity.
