Effect of 25 (OH) vitamin D3 on urinary excretion of cyclic adenosine monophosphate

The fractional excretion of P (P clearance/inulin clearance) and the excretion of cyclic AMP (cAMP) in urine were estimated for intact female Sprague-Dawley rats given by vein 25-hydroxycholecalciferol 0.1 mu g/100 g h, parathyroidectomized (PT) rats given by continuous infusion parathyrin 1 U/100 g h and PT rats given parathyrin and 25-hydroxycholecalciferol. In the 3 groups fractional excretion of P fell from a control value of 0.210 to 0.052, rose from 0.031 to 0.365 and fell from 0.365 to 0.256, respectively. In urine cAMP fell from 182 to 87, rose from 76 to 330 and fell from 356 to 191 pmol/min. The results suggest that the mechanism underlying the increased tubular reabsorption of P is inhibition of the activation of adenylate cyclase in kidney induced by parathyrin.
