Clinical trial of pyrantel embonate, levotetramisole and bephenium hydroxynaphthoate against Necator americanus

Pyrantel embonate (Combantrin), levamisole (Decaris) or bephenium hydroxynaphthoate (Alcopar) were given orally as single doses at the recommended levels to 73 fasting patients in Singapore infected with Necator americanus. 66 of them completed the follow-up (faecal examinations 7 and 14 days after treatment); of these 20 also had Ascaris, 26 Trichuris and 14 both Ascaris and Trichuris. By the 2nd faecal examination pyrantel embonate had cleared N. americanus from 16 of 20 with less than 5,000 e.p.g. and 2 of 3 with 5,000 to 15,000 e.p.g. Similarly, levamisole cleared 13 of 19 and none of 2, respectively, and bephenium hydroxynaphthoate cleared 7 of 16 and one of 6. All 3 anthelmintics were also effective against Ascaris, but none was effective against T. trichiura. More side effects were observed with bephenium hydroxynaphthoate than with the other 2 anthelmintics.
