Pathophysiological investigations of Schistosoma mansoni infection in Mastomys natalensis under chemotherapy with a nitrovinylfuran derivative (SQ 18.506)

The nitrovinylfuran derivative trans-5-amino-3-[2-(5-nitro-2-furyl)-vinyl]-1,2,4 oxadiazole (SQ 18,506) showed 97.9% activity against Schistosoma mansoni in Mastomys natalensis, when given orally at 500 mg/kg body-weight on 5 consecutive days. The egg output was also markedly reduced after the administration of subcurative doses (5 X 250 mg/kg). Treatment of uninfected animals with the minimum curative dose revealed no side effects or host toxicity. Pathophysiological studies revealed a peak of SDH activity 6 to 7 weeks after infection; in treated animals a further peak occurred about 2 weeks after the beginning of treatment. The levels of leucocytes and eosinophilic granulocytes showed sharp but transient maxima at 7 weeks after infection; treatment had little effect on these values.
