For 5 days rats were given subcutaneously ergocalciferol 25 000 or 50 000 IU/g bodyweight daily without or with saline 3 ml/g intraperitoneally and were killed on day 6. Others had 50 000 IU/g for 5 days and were killed 31 days after the last dose, 25 000 IU/g for 15 days or 0.2 ml parathyroid extract for 5 days. In rats given 25 000 IU ergocalciferol plasma Ca rose from 9.87 to 12.48 mg/100 ml and Mg fell from 1.85 to 1.58 mg/100 ml; in those on 50 000 IU, Ca rose to 16.83 and Mg to 2.88 mg/100 ml. In those given saline as well as ergocalciferol plasma Ca:Mg ratio became linear and values were inversely related. In all other groups plasma Ca rose and Mg fell. It is suggested that hypermagnesaemia in acute hypercalcaemia is secondary to renal failure; hypomagnesaemia of the other groups is a direct consequence of hypercalcaemia which is exaggerated in those given saline because of the prevention of uraemia and the production of Na diuresis.
