Fluorescence serological and histological studies on CELO-virus infected chick embryos

After injection into chick embryos on the tenth day of incubation, the Phelps strain of CELO virus was first detectable in endodermal cells of the chorioallantoic membrane (CAM) at 10 hours, irrespectve of the route of inoculation (CAM, allantoic cavity, amniotic sac), and the fluorescence was present, first in the nucleus, then in the cytoplasm, up to death of the embryo within 48-84 hours. Viral antigen was not formed elsewhere except after inoculation into the allantoic cavity, when it also appeared in cells of the lung, kidneys, intestinal and nasal mucosa, and the feather marrow. Histological lesions were in all cases confined to the CAM, and two types of inclusion body were distinguished: eosinophilic, Feulgen-positive bodies of Cowdry type A, and basophilic, Feulgen-negative bodies not typable in the Cowdry scheme. Since the endodermal lesions in the CAM appeared to be an insufficient cause of death in themselves, it is suggested that they increase the permeability of the CAM to toxic metabolic products (e.g. uric acid), thereby exposing the chick to autointoxication.
