A serum-neutralization test for canine distemper virus in cell culture in microtitration plates was found to be as sensitive as the conventional test in chick embryos. The results of 148 samples indicated that the microtest had either greater sensitivity or smaller measurement error. Reproducibility of serum titres using the microplate assay was analysed. The combined effect of all sources of variability within a laboratory resulted in a standard error of plus or minus 0.3 (log10) for a sample of serum titrated at threefold dilutions with 4 replicates per dilution.
