Metabolism of sulphonamides in goats

After initial i/v injection followed by continuous i/v infusion of sulphanilamide, sulphadimidine, sulphamethoxazole or sulfadoxine, these sulphonamides were excreted mainly unchanged in the urine of goats. The unchanged drug amounted to 33% for sulphadimidine, 52% for sulphanilamide, 82% for sulphamethoxazole and 42-62% for sulfadoxine. The respective figures for the N4 acetylated drug in the urine were 4, 38, 13 and 22-27%, and for conjugated metabolites 30, 7, 5 and 16-31%. The range of plasma concentrations for sulphadimidine was 31-180 mu g/ml, sulphanilamide 27-243, sulphamethoxazole 30-171 and sulfadoxine (in two series of experiments) 46-86 and 186-233 mu g/ml. An increase in the plasma concentration of sulfadoxine was followed by a relatively higher concentration of the unchanged drug in the urine.
