Octylonium bromide was tested for its spasmolytic effect on a number of in vitro preparations involving the gastrointestinal tract front man, guinea pig and rabbit. In all the different experiments the compound showed marked inhibitory activity on both basal motility and motility induced by physiologic spasmogenic agents like acetylcholine, histamine and peptides acting directly on smooth muscle (substance P and its analogues). On the guinea pig ileum the inhibitory action appeared to be of the competitive type against acetylcholine and non-competitive against histamine and peptides suggesting a predominant anticholinergic effect. However in all the other preparations the inhibitory action appeared to be myotropic in origin as was observed in in vivo preparations. Quantitative differences in the various preparations were probably connected with different sensitivity to the various stimulatory agents.
