Octylonium bromide was tested for its spasmolytic activity on a number of in vivo preparations involving different animal species. Also the possible interference with gastric secretion was investigated. The compound appeared to be devoid of inhibitory effects on both basal and stimulated gastric secretion, when administered orally or intravenously. The spasmolytic effect was present in all the various experimental conditions though with consistent quantitative differences: guinea pig gall bladder was particularly sensitive whereas the rat pyloric sphincter was relatively insensitive. A remarkable relaxant effect was observed in the rat stomach with a consequent delay in gastric emptying: however this phenomenon could be noted only after intraperitoneal but not oral administration of octylonium bromide. With the only exception of a delay in gastric emptying probably referable to its anti-muscarinic properties, octylonium bromide exerts its spasmolytic activity mainly through a direct myolitic effect.
