Two different chromatographic techniques have been developed for the determination of ketamine in the plasma of humans. The first uses a flame ionization technique and permits only the quantification of unchanged ketamine. The second uses an electron capture detector and makes possible detection of unchanged ketamine and its main metabolites in the form of heptafluorobutyric anhydride. The variation coefficient of the flame ionization technique was 8.5% and the degree of extraction recorded was 99.45%, while the electron-capture detector gave a variation coefficient of less than 6%, both for ketamine and its two derivatives. The latter method was used to determine the pharmacokinetic parameters of ketamine and its two metabolites in man.
