DMO and nicotine have been extensively used for the determination of intracellular pH using the principles of non-ionic diffusion. Ionization constants (expressed in pKa) of these two substances were obtained over a wide range of temperature and salinity going from those of fresh water to those of sea water including appropriate conditions for mammalian fluids. A potentiometric titration was used to measure the pKa values for DMO and nicotine. This work offers the possibility of selecting pKa values of these two substances at any biological condition using a graphical method. It stresses the importance of knowing the ionization constant values when non ionic diffusion is used for intracellular pH determinations.
