The rate of epimerization of tetracycline under various experimental conditions was investigated. It appears that the addition of solubilizers leads to a significant decrease of the reaction rate. In case of urea and polysorbate 20, the stabilizing effect is generally less than that obtained by the presence of thiourea and PEG 6000. For urea, this could be explained by the fact that urea, by altering the water structure, would weaken the intermolecular hydrophobic interactions. For polysorbate 20, this fact should be accounted for the presence of trace metal impurities in the commercial grade surfactant used. Similar rate pH profiles were observed in the presence and absence of solubilizers. Data obtained at various temperatures permitted calculation of the activation energy of the reaction and prediction of the half lives of tetracycline in the different systems, at room temperature.
