1. Bone-rudiments from seven day chick embryo were cultivated by watch glass method and the effect of progesterone (1.25 mg./0.125 mg. and 0.0125 mg./ml. medium) on such rudiments was studied. 2. Progesterone had a definite and progressive retarding effect on the growth of the rudiments in accordance with the concentrations of hormone used. 3. Withdrawal of progesterone from the medium after two days cultivation showed no further reduction in their growth and a growth rate similar to control was regained showing that the inhibitory action of the hormone was temporary and will be lost on cessation of treatment. 4. The rudiments reacted more intensely to hormone when embryo extract was withdrawn from the medium. 5. Histological study revealed more vacuolization of the hypertrophic cells and reduction in matrix. 6. Progesterone had a direct inhibitory action on bone growth in vitro and reduction in length may be due to its action on matrix similar to that of vitamin A.
