Orally administered granisetron has equivalent antiemetic efficacy to granisetron administered by the intravenous route and, in animal models, is at least as potent. The principal sites of action of granisetron are 5-HT3 receptors located on the vagal afferent neurones adjacent to the enterochromaffin cells in the gut mucosa. Absorption of granisetron occurs across the duodenal mucosa. Granisetron exhibits a very high receptor specificity, strong potency, good efficacy, and a curvilinear dose response curve. Granisetron's long duration of action makes it an excellent choice for the control of acute-phase emesis induced by chemotherapy in cancer patients.
