An investigation is described to determine a satisfactory method of killing and preserving nematodes in soil for later extraction by sugar flotation. Fumigation of soil samples with methyl bromide followed by freezing at -15 degrees C was satisfactory and gave good recovery of Meloidogyne incognita larvae, Tylenchorhynchus claytoni and Helicotylenchus dihystera, but was time consuming. The effectiveness of microwave treatment was influenced by soil volume, container and soil moisture.
