The results of author's own studies and data from the literature were systematized. These data demonstrate the presence of a direct correlation between drug tolerance and changes in the functional state of pineal body. The disengagement of the pineal body decreases drug tolerance, but the administration of melatonin, the most important pineal hormone, intensifies adaptation to drugs. Possible mechanisms of these phenomena were considered.
