Effect of thiol protectors on cell cycle phases in murine bone marrow

Effect of S-2-(3-aminopropylamino) ethyl phosphorothiotic acid (WR-2721, 200 mg/kg), and 2-mercaptopropionylglycine (MPG, 20 mg/kg) on the cell cycle progression in mouse bone marrow was studied by flow cytometry. The drugs were injected (ip) to adult Swiss albino mice either individually or in combination and DNA distribution in the bone marrow cells was measured from 1 to 30 hr post-injection. The results showed an immediate G-2 block in all the treated groups but the major effect of WR-2721 was on G-1 phase resulting in a cell cycle delay. MPG appears to accelerate the cell cycle after initial G-2 block. In combination, the individual effects of both the drugs were fully manifested. The cell cycle response of in vivo systems like mouse bone marrow may be influenced by the microenvironment and the heterogeneity of the cell population.
