A double-blind, proctoscopic and clinical comparison of flurbiprofen suppositories and ibuprofen suppositories in osteoarthrosis

A double-blind, parallel-group, comparative study of flurbiprofen 100 mg suppositories and ibuprofen 500 mg suppositories, 1 of either suppository being used morning and night for 20 days, was done on 30 patients with osteoarthrosis. Flurbiprofen and ibuprofen achieved a highly significant reduction in arthritic pain at rest during their 20 days of usage (P < 0.004). There was no significant difference between the analgesic activity of flurbiprofen and ibuprofen (P > 0.05). There was a reduction in the mean duration of morning stiffness and a minimal improvement in the mean score of grip strength of both the flurbiprofen and ibuprofen treatment groups during the 20 days of study. Laboratory investigations including Hb, erythrocyte sedimentation rate (ESR), proteinuria and serum levels of glucose, transaminase bilirubin, alkaline phosphatase, uric acid, urea and cholesterol were unchanged at completion of the study. Three patients were withdrawn from the study, the first being a patient on flurbiprofen who developed diarrhea at day 14 of the study. The other 2 withdrawn patients were on ibuprofen, one developed diarrhea and anal burning sensation at day 12, the other a generalized urticaria at day 7 of the study. Proctoscopic examination of the remaining 27 patients in the study as performed at day 0 (before treatment was commenced), day 10 and day 20 (on completion of treatment). All 27 patients had good local anorectal tolerance to the trial suppositories. In 50% of the cases treated with flurbiprofen suppositories, there was an improvement in proctoscopic appearances on completion of trial. A photographic record of the proctoscopic appearance of some patients on entry to and completion of the study was made.
