Antiulcer activity of sodium valproate in rats and guinea pigs

The effect of sodium valproate (sod. valproate) and cimetidine was studied on gastric secretion and ulceration induced by 2 hr restraint at 4.degree. C, forced swimming (5 hr at 33 .+-. 1.degree. C) or pylorus ligation in albino rats and histamine induced acute gastric and duodenal ulceration in guinea pigs. Sod. valproate administration (po) significantly reduced the gastric ulceration induced either by 2 hr restraint at 4.degree. C or forced swimming in a dose dependent manner similar to cimetidine. Both sod. valproate (400 mg/kg, po) and cimetidine (50 mg/kg, po) significantly reduced the gastric secretory volume, total and free acidity and peptic activity. They also prevented ulcers induced by pylorus ligation. Sod. valproate (400 mg/kg, po) significantly reduced the gastric ulceration but was ineffective against duodenal ulceration induced by histamine. On the contrary, cimetidine (50 mg/kg, po) prevented both the gastric and the duodenal ulceration.
