3-acetylpyridine 3-AP, an analogue of nicotinamide(NA)leading to formation of the 3-AP analogue of DPN, was administered subcutaneously to rats. It was found that LD50 was 92 mg./kg./24 hours or 75 mg./kg/l4 days. Metabolic and endocrine changes were studied following the administration of a sublethal dose(LD10)of 3-AP, i.e. 60 mg./kg., to male rats. It was found, 1) that food intake was greatly diminished for several days, resulting in a marked decrease in weight gains, 2.) that the urine flow, after the administration of the same dose of 3-AP, underwent no marked changes and that even the excretion of a water load administered several days after acute poisoning was not affected, 3) that the administration of 3-AP was first followed by several hours hypoglycaemia, which later changed to hyperglycaemia lasting 1-2 days, 4) that no marked changes occurred in the weight of the endocrine glands up to 14 days after the administration of 3-AP, 5) that 3-AP did not affect the capacity of the hypophysis to react to unilateral adrenalectomy by over- production of ACTH or to a methylthiouracil block of the thyroid by overproduction of (TSH) thyrotropin.
