Synthesis of prostanoids in the gastric and duodenal mucosa of healthy subjects

Biopsy specimens of 6 normal subjects taken from stomach (body and antral areas) and duodenal bulb during endoscopy were utilized for prostanoid synthesis in short term culture in absence (BR) and in presence (SR) of calcium ionophore (A23187). The higher ability of body mucosa to synthesize prostanoids (PGE2, TXB2) in absence of calcium ionophore is probably due to the different cellular composition. PGE2 and PGD2 synthesis decrease significantly during SR, while 6KPGF1.alpha. (at body level) and TXB2 (at duodenal bulb level) is significantly increased. These results may suggest that: 1) mucosal epithelial cells are the major source of PGE2, PGD2 and PGF2.alpha., while thrombocytes, endotelial cells and monocytes synthesize especially TXB2 and 6KPGF1.alpha.; 2) the lower ability of antral and duodenal mucosa to produce prostanoids, besides other factors (e.g. antral stasis, duodenal acid load) can explain the major inflammatory susceptibility of these areas.
