Effect of megestrol acetate on the genital organs & fertility of male rats

Oral administration of megestrol acetate (17ɑ-acetoxy-6-dehydro-6-methylprogesterone; 40 mg/kg body wt/day for 30 days) has no effect on spermatogenesis and fertility of rats. However, the weight of the genital organs is reduced and the gonadotropin content of the pituitary is elevated. These changes are reversed upon discontinuation of treatment. Megestrol acetate antagonizes the action of exogenous androgen on the accessory genital organs of castrated rats. The significance of these findings is discussed.
