Monoclonal antibodies against erythrocytic stages of Plasmodium vivax inhibitory effect on gametocyte infectivity and development within mosquitoes

Using hybridoma technique, a panel of monoclonal antibodies against erythrocytic stages of Plasmodium vivax has been produced and reported elsewhere. In order to identify whether these McAbs have protective significance, we chose five of them with broad-spectral IFA reactions (1D1-G7; 6H7; 7A7; 6F7 and 1D6-H10) for this study. McAbs were fed to Anopheles stephensi together with the patient's gametocyte-containing blood through the membrane feeders. The effect of the test McAbs in the blood meal was compared with that of both positive and negative controls and measured by subsequent examination of the mosquitoes for the presence of oocysts and sporozoites. Six experiments were successfully carried out dueing two epidemic seasons in 1986-1987. Results of these membrane feeding experiments demonstrated that four McAbs inhibited gametocyte infectivity to different extent as compared with the negative controls. Among them, 1D6-G7, a subclone, was shown to be the most effective one. Under the same experimental conditions, the percentage of mosquitoes infected in 1D6-G7 , chloroquine, SP2/0 ascites and untreated groups was 20.0%, 60.0%, 70.0% and 57.7% respectively; the average number of oocysts per positive gut dissected in the corresponding groups was 4.1 (P<0.01) 13.8, 38.5 and 24.7 respectively. According to the mean number of oocysts, the inhibitory rate induced by 1D6-G7 was 84.6%. Since McAb 1D6-G7 has apparently suppressive effect on gametocyte infectivity and its development within mosquito vectors shown above, it may play an important role in the further studies on the transmission-blocking immunity in Plasmodium Vivax.
