Experimental study of binding and effects of cardiac glycosides in the coronary circulation of the intact and ischemic heart

The paper presents new evidence of the vasoactivity cardiac glycosides in the myocardium. In experiments performed on the in situ dog heart, by utilizing hemodynamic measurements (1), Romhanyi's polarization microscopic topoptical (ABT) method adapted to the demonstration of glycoside binding (2), and cardiac telethermography (3), respectively, a close functional and morphological parallelism has been documented the strophantoside induced coronary actions. The strong binding of strophantoside to the coronary vessel wall and the endothelial cells was found to be accompanied by vasoconstriction both in the intact and the regionally ischemic heart muscle. Local decrease of blood flow as observed within the ischemic area can be explained by an enhanced binding of the drug to the border zones which supply collateral channels. The potential clinical impact of these findings on therapy is briefly evaluated. The non-qualitative methods are useful for investigation of glycoside binding.
