Loading and maintenance dose for the determination of amino acid kinetics in plasma

While fasting 8 metabolically healthy persons had by vein a loading dose of amino acid mixture (Thomaeamin n 10%Reg.) at 20 mg/kg min for 5 min and a maintenance dose at 5 mg/kg min for 55 min. Plasma amino acid concentrations were estimated by Biotronic LC 6001 before, during and for 120 min after infusion. Mean distribution volume of individual amino acids was from 4.7 to 9.4 litres, elimination rate constant from 1.7 to 11.0/h, total clearance 186 to 846 ml/min and transfer or endogenous production rate 12 to 135 mu mol/kg h. Total transfer reached 17.6 mmol, equivalent to 2.2 g amino acids, per kg daily. An experimental design permitting increase in plasma amino acid concentration to 2 to 3 times the starting value, use of a test mixture adapted to transfer rate of the subjects and calculation of kinetic indices from a 2-compartment model, or without a model, are recommended.
