A single reagent method of phosphate estimation in phosphatase (s) assay

A simple and sensitive method of phosphorus estimation suitable for phosphatase(s) assay is described. The method is based on the formation of protein-phosphomolybdate complex under relatively mild acidic conditions in the presence of a non-ionic detergent and low molybdate content. The complex has a .lambda.max at 318 nm with an .epsilon. of 2.31 .times. 104 and compares reasonably well with the existing methods. The relative stability of some of the phosphoester substrates under the assay conditions and the adoption of the proposed method for phosphatase(s) assay are described.
