Crystal violet ring response as a marker for in vitro detection of pathogenic Vibrio parahaemolyticus & Shigella spp

A total of 311 strains belonging to 11 genera and 34 different species was studied to see the reaction patterns of different bacteria with respect to crystal violet (CV) ring formation as an aid to rapid diagnosis of pathogenicity (in vitro). It was found that only the Vibrio parahaemolyticus strains, at 75 or 100 .mu.g/disc concentration, could produce 'double ring (DR) response which was different from that of all other bacteria tested, except Staphylococcus aureus. 'Single rings' (SR) were produced by some V. cholerae, V. alginolyticus, Providencia and many S. aureus strains. Some enterotoxigenic strains of Shigella spp. and Escherichia coli also produced a type of dense ring, resembling somewhat the DR response of V. parahaemolyticus, which, however, merged with the disc itself unlike that of the former two. The DR type of CV response was found to have an absolute and specific correlation with enterotoxigenicity of the V. parahaemolyticus strains only, like that of S. aureus.
