Binding of [superscript 3H]dexamethasone to specific cytosol binding sites of rat mammary gland during pregnancy and lactation was characterized and quantified. During the first wk of pregnancy, amount of receptor gradually increased and then remained at a relatively constant value throughout pregnancy. Max. receptor concn. were observed immediately before and 1 day after parturition. During lactation, receptor concn. gradually returned to the value observed during pregnancy and then remained relatively constant until day 21 after which there was a rapid decrease to end of lactation. No significant differences in dissociation constants were noted during pregnancy and lactation, values ranging from 10 to 30 nM.
