Activities of fatty acid synthetase, palmitoyl-CoA synthetase and glycerol-3-phosphate acyl transferase in rabbit mammary gland were low in non-pregnant, non-lactating animals and increased slightly during pregnancy. Following parturition, enzyme activities had increased 4-5-fold by wk 1 post partum, followed by a gradual decrease from wk 2. Studies on substrate specificity of glycerol-3-phosphate acyl transferase demonstrated a preference for palmitoyl-CoA over shorter chain acyl-CoAs. Mammary gland medium-chain thioesterase activity was not detectable in the non-pregnant, non-lactating rabbit, but appeared in the gland during pregnancy at an activity which was maintained throughout lactation.
