A new monoclonal antibody GRV1 is presented. This monoclonal antibody was produced immunizing with the peripheral mononuclear cells of a patient diagnosed of a prolymphocytic leukaemia (Ia/DR positive, IgM positive K). After four weekly invections of 107 cells, spleen cells of BALB/c mice were fused with NS1 myeloma, GRV1 monoclonal antibody was characterized by indirect immunofluorescence against a panel of normal cells, fifteen human cell lines of different etiology and origin as well as forty six leukaemias. GRV1 reacted only with the immunizing cells and it was demonstrated in capping experiments that bound the surface immunoglobulin of the prolymphocytic leukaemic cells. Immunoprecipitation SDS-PAGE analysis and autoradigraphy confirmed that GRV1 precipitated two peptides of 70 and 23 KD respectively, that correspond with the molecular weight of the heavy and light chain of the IgM. We conclude that the monoclonal antibody GRV1 probably recognize the idiotype of the surface IgM of the prolymphocytic leukaemia.
